چکیده

The medium composition was as follows : 0.15 M-sucrose, 30rn~-KCl, 1OmM-choline chloride, 10mM-succinate/Tris, lOmM-Tris/Mops, 3mM-P,/Tris, 2pM-rotenone, 0.1 mMEGTA and 2mg of rat-liver mitochondria/ml; pH7.4. Temperature 25°C. Respiration was stimulated by 0.5 m ~ ADP ( 0 ) in the presence of 1 mM-EDTA, by 20pmol of FCCP/mg (0 ) or by 6Opmol of FCCP/mg (0). When the respiration was stimulated by ADP or by 6Opmol of FCCPlmg, malonate at a concentration between 0 and 5 m ~ was used as inhibitor. When respiration was stimulated by 20pmol of FCCP/mg the inhibitor was antimycin at a concentration between 0 and 12pmol/mg.

Protonmotive force as the source of energy for adenosine 5'-triphosphate synthesis in Escherichia coli.

Net synthesis of adenosine 5'-triphosphate (ATP) in energy-depleted cells of Escherichia coli was observed when an inwardly directed protonmotive force was artificially imposed. In wild-type cells, ATP synthesis occurred whether the protonmotive force was dominated by the membrane potential (negative inside) or the pH gradient (alkaline inside). Formation of ATP did not occur unless the protonm...

Thyroid-hormone control of state-3 respiration in isolated rat liver mitochondria.

Oxidative phosphorylation can be treated as two groups of reactions; those that generate protonmotive force (dicarboxylate carrier, succinate dehydrogenase and the respiratory chain) and those that consume protonmotive force (adenine nucleotide and phosphate carriers. ATP synthase and proton leak). Mitochondria from hypothyroid rats have lower rates of respiration in the presence of ADP (state ...

Adenosine 5'-triphosphate synthesis driven by a protonmotive force in membrane vesicles of Escherichia coli.

Adenosine 5'-triphosphate (ATP) synthesis energized by an artificially imposed protonmotive force (delta p) in adenosine 5'-diphosphate-loaded membrane vesicles of Escherichia coli was investigated. The protonmotive force is composed of an artificially imposed pH gradient (delta pH) or membrane potential (deltapsi), or both. A delta pH was established by a rapid alteration of the pH of the assa...

ATP is essential for protein translocation into Escherichia coli membrane vesicles.

The energy requirement for translocation of alkaline phosphatase and the outer membrane protein OmpA into Escherichia coli membrane vesicles was studied under conditions that permit posttranslational translocation and, hence, prior removal of various components necessary for protein synthesis. Translocation could be supported by an ATP-generating system or, less well, by the protonmotive force ...

Coupling of proton flow to ATP synthesis in Rhodobacter capsulatus: F(0)F(1)-ATP synthase is absent from about half of chromatophores.

F(0)F(1)-ATP synthase (H(+)-ATP synthase, F(0)F(1)) utilizes the transmembrane protonmotive force to catalyze the formation of ATP from ADP and inorganic phosphate (P(i)). Structurally the enzyme consists of a membrane-embedded proton-translocating F(0) portion and a protruding hydrophilic F(1) part that catalyzes the synthesis of ATP. In photosynthetic purple bacteria a single turnover of the ...
